Rad51-based metabolic disease therapeutic that enhances muscle glucose uptake by binding to AMPK

This technology is a pharmaceutical composition for preventing or treating one or more metabolic diseases, including diabetes, obesity, hyperlipidemia, fatty liver, and hypertension, using Rad51 protein or a Rad51-expressing vector as an active ingredient. It also includes food products for prevention and improvement containing the same protein, as well as a screening method for identifying therapeutic candidates that enhance Rad51 activity or expression.

Many patients with type 2 diabetes struggle to reach their HbA1c targets due to difficulties in blood glucose control. While widely used incretin hormones are rapidly degraded by DPP-4, leading to the development of DPP-4 inhibitors and GLP-1 analogs, these have shown limited efficacy or have been associated with risks of pancreatitis and pancreatic cancer. Although AMPK, an energy sensor, has emerged as a key target for treating insulin resistance, there has been a lack of novel regulators to activate it.

In this technology, Rad51 directly binds to AMPK in muscle tissue to promote AMPK phosphorylation, which increases muscle glucose uptake and inhibits fatty acid synthesis through ACC phosphorylation. The screening method involves treating Rad51-expressing cells with candidate substances and selecting those that increase Rad51 activity or expression compared to a control group. This can be applied to the development of new type 2 diabetes drugs, functional foods for obesity and fatty liver, and AMPK pathway drug screening, offering a novel therapeutic strategy that targets both blood glucose reduction and inhibition of fat synthesis through a single protein.
