This technology protects neurons by treating glutamate-damaged HT22 cells or amyloid-beta-induced PC12 cells with extract from the brown alga *Sargassum horneri* (pae), serving as a foundation for developing materials to prevent and treat degenerative brain diseases.
Excessive reactive oxygen species in the body cause oxidative stress, damaging proteins, lipids, and DNA, which are major causes of central nervous system degenerative diseases like Alzheimer's, Parkinson's, and Huntington's. While *Sargassum horneri* is known for antioxidants like fucoxanthin and phlorotannin, its neuroprotective efficacy and mechanisms were previously unknown.
This technology utilizes the extract's ability to inhibit cell death in glutamate-damaged cells by increasing Akt phosphorylation, lowering the Bax/Bcl-2 ratio and caspase-3 expression, and increasing HO-1/Nrf2 expression. In amyloid-beta-damaged cells, it reduces inflammatory responses by lowering MAPKs phosphorylation. The extract is obtained using lower alcohols, dried, and dissolved in 1,3-butylene glycol. It can be applied to dementia prevention research, neuroprotective screening, and brain health product development, enabling the use of native coastal brown algae as a natural neuroprotective ingredient targeting both oxidative damage and neuroinflammation.
