This technology provides pharmaceutical and food compositions for promoting pregnancy and preventing or treating infertility, using chemokine CCL23 as an active ingredient. It also includes methods for enhancing endometrial cell proliferation in non-human mammals.
With declining pregnancy rates due to advanced maternal age and increased stress, implantation-related infertility that remains unresolved even by IVF is a major challenge. Successful implantation requires sufficient endometrial cell proliferation and differentiation, as well as a reduction in endoplasmic reticulum stress—a cause of placental abnormalities and miscarriage—but no substance has previously met both conditions.
This technology leverages the ability of CCL23 to activate protein expression within the PI3K/AKT and ERK1/2 MAPK signaling pathways to boost endometrial cell proliferation, while simultaneously reducing endoplasmic reticulum stress-inducing proteins such as IRE1α, ATF6α, p-PERK, p-eIF2α, GRP78, and GADD153. It addresses a wide range of infertility issues, including implantation failure, anovulation, and tubal, uterine, or cervical infertility. It can be applied as an endometrial preparation supplement for IVF, a dietary supplement for women with recurrent implantation failure, and a technology to improve embryo transfer success in high-quality livestock, thereby optimizing the uterine environment for implantation and increasing pregnancy success rates.
This invention was developed with support from the Ministry of Agriculture, Food and Rural Affairs for the development of early selection indicators and the enhancement of reproductive/immune capacity in high-parity sows to improve lifetime productivity.
