This technology is a pharmaceutical composition for the prevention and treatment of insulin resistance-specific type 2 diabetes, and a health functional food for prevention and improvement, containing biglycan—a myokine protein secreted from skeletal muscle—as an active ingredient. Biglycan lowers blood sugar by increasing AMPK phosphorylation and cellular glucose uptake.
Type 2 diabetes is difficult to manage, with only about 40% of patients reaching their target glycated hemoglobin levels due to challenges in blood sugar control. Widely used incretin-based therapies suffer from the rapid degradation of GLP-1 and GIP by DPP-4, and insulin treatment often has limited efficacy because many cases are caused by signaling abnormalities downstream of the insulin receptor.
This technology is configured to promote glucose utilization by increasing the expression and activity of AMPK, an energy sensor in metabolic tissues, using biglycan represented by the amino acid sequence of SEQ ID NO: 3, effectively implementing the blood sugar-improving effects of exercise-induced muscle substances as a protein-based material. It can be applied to insulin-resistant diabetes protein therapeutics, health functional foods for managing hyperglycemia caused by lack of exercise, and the discovery of new AMPK-targeted metabolic disease drugs. This provides a new pathway to lower blood sugar in patients with insulin signaling abnormalities while overcoming the short duration limitations of incretin agents.
This invention was developed with support from the Ministry of Science and ICT for the discovery of novel myokines and the characterization of their roles in metabolic diseases.
