This technology is a pharmaceutical and food composition for preventing, improving, and treating skeletal muscle atrophy, containing one or more of kukoamine A, kukoamine B, or their pharmaceutically acceptable salts as active ingredients. The active ingredients can be extracted and isolated from Lycii Cortex or potato peels.
Muscle mass loss is caused by aging, lack of exercise, pathological conditions like cachexia or chemotherapy, and excessive exposure to stress hormones, leading to reduced walking speed, falls, fractures, and metabolic diseases. Specifically, glucocorticoids inhibit the PI3K/Akt/mTOR pathway to block muscle protein synthesis and activate degradation genes like Atrogin-1 and MuRF-1. However, previous research on kukoamine was limited to neuroprotection or fatty liver improvement, with no known application for muscle atrophy.
This technology is configured for kukoamine to lower the expression of muscle atrophy proteins such as Atrogin-1, MuRF-1, Myostatin, and SIRT1, while activating muscle synthesis proteins like PI3K, Akt1, and TRPV4, as well as mTOR-based protein synthesis mechanisms. It also inhibits muscle fiber bundle reduction, muscle inflammatory cell infiltration, and local fibrosis. Extraction can be performed by adding edible organic acids to water or lower alcohols. It can be applied to health supplements for age-related sarcopenia, aids for disuse atrophy in long-term bedridden or casted patients, and nutritional products for cachexia management during chemotherapy, providing a material that regulates muscle degradation and synthesis using discarded potato peels and traditional Lycii Cortex.
This invention was developed with research support for natural materials to improve skeletal muscle loss from the Ministry of Science, ICT and Future Planning.
