This technology involves a pharmaceutical composition and health functional food for pain suppression and prevention, containing BigLEN (SEQ ID NO: 1 or 2, e.g., LENPSPEAPARRLLPP), a brain-abundant neuropeptide, or its pharmaceutically acceptable salts as an active ingredient. BigLEN exhibits analgesic effects that suppress hyperalgesia induced by various stimuli.
Current analgesics are largely divided into non-steroidal anti-inflammatory drugs (NSAIDs)/COX-2 inhibitors and opioids like morphine, both of which have shown little efficacy for neuropathic pain. Opioids show some effect only at high doses but carry risks of severe side effects and addiction, while NSAIDs require higher doses for efficacy, leading to gastrointestinal side effects.
This technology is based on the confirmation that the L-amino acid-based BigLEN peptide reduces pain in models of formalin-induced pain, CFA-induced inflammatory hyperalgesia, mechanical/thermal hyperalgesia, and neuropathic pain. Target conditions include postoperative pain, cancer pain, chemotherapy-induced pain, postherpetic neuralgia, fibromyalgia, and migraines. It can be applied to chemotherapy-induced peripheral neuropathy analgesics, non-narcotic injections for postoperative pain, and health supplements for chronic pain, addressing neuropathic pain that existing drugs cannot treat without the addiction risks of opioids or the GI issues of NSAIDs.
This invention was developed with support from the Ministry of Science and ICT for the development of core technologies for pain control and assessment based on input neural network molecular mechanisms.
