Oligoamine Boronic Acid for Heparin Receptors

Listed on

Material for Intraoperative Blood Heparin Measurement and Removal Using Oligoamine Boronic Acid Receptors

This technology is a heparin-binding composition featuring oligoamine boronic acid (OAB) as an active ingredient, containing cationic amine groups and 1 to 3 phenylboronic acid groups. As a biodegradable synthetic receptor based on natural amino acids and 3-carboxyphenylboronic acid, it binds selectively and strongly to heparin under neutral conditions.

Heparin, used as an anticoagulant in cardiopulmonary bypass surgery, requires continuous blood concentration monitoring to prevent bleeding complications; however, conventional activated clotting time (ACT) measurements are limited by being indirect indicators. Furthermore, existing boronic acid-based receptors have had difficulty in practical blood applications due to low heparin-binding affinity at physiological neutral pH.

This technology utilizes both ion pairing of amine groups and boronic acid ester formation to achieve a heparin-binding affinity over 150 times higher than conventional boronic acid receptors. When combined with a pyrocatechol violet indicator, blood heparin concentration can be read directly through color changes. It can be applied to intraoperative anticoagulation monitoring kits and postoperative heparin neutralizers, simplifying hemostasis management and improving patient recovery.

Key Features:
  • A heparin-binding composition containing as an active ingredient an oligoamine boronic acid of Formula 3, wherein at least one of R1, R2, and R3 is a phenylboronic acid carbonyl group.
  • A heparin-binding composition with a heparin binding constant increasing from 9.78×10^5 to 5.82×10^6 M-1 depending on the number of phenylboronic acid groups.
  • A heparin-binding composition that quantifies intraoperative blood heparin levels and allows direct concentration verification via colorimetry using pyrocatechol violet as an indicator.
  • A biodegradable composition that binds strongly to heparin at neutral pH 7.4 and is used as a blood heparin remover after surgery.
